Zu seinen bekanntesten Rollen gehörten die Titelfigur in Dracula (1958) sowie neun weiteren Vampir-Filmen, Francisco Scaramanga im James-Bond-Film Der Mann mit dem goldenen Colt (1974), Saruman in Der Herr der Ringe (2001-2003) und Der Hobbit (2012-2014) sowie Count Dooku in Star Wars (2002, 2005). Außerdem wirkte er in fünf Filmen von Tim Burton mit, unter anderem auch in Dark Shadows (2012). Zuletzt war er in dem Finale der Trilogie Der Hobbit zu sehen. Lee wurde trotz seiner ikonischen Rollen nie für einen Oscar nominiert. Als seine wichtigste Arbeit bezeichnete Lee die Darstellung des Politikers Muhammad Ali Jinnah im 1998 erschienenen Film Jinnah; er gab den Film ebenfalls als seine beste schauspielerische Darbietung an. Seit 1946 hatte er in über 285 Filmen mitgewirkt. Neben der Schauspielerei war Lee auch als Sänger aktiv. So wirkte er zwischen 1986 und 1998 bei Studioaufnahmen verschiedener Opern und Musicals mit und veröffentlichte 2010 das Symphonic-Metal-Album Charlemagne: By the Sword and the Cross, nachdem er bereits seit 2005 mit Bands wie Rhapsody of Fire und Manowar zusammengearbeitet hatte.
